DSIP — Delta Sleep-Inducing Peptide / Neuropeptide Sleep & Stress Modulator

DSIP is a synthetic nonapeptide — nine amino acids in sequence (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) — first isolated in 1974 from the cerebral venous blood of sleeping rabbits by Swiss researchers at the Schoenenberger-Monnier group. Since its discovery, it has become one of the more widely studied endogenous neuropeptides in sleep research, with a biological profile that extends well beyond sleep architecture into stress hormone regulation, neuroprotection, and neuromodulation. Its multi-system activity and naturally occurring origin make it a compelling subject in neuroscience and CNS research.

Why DSIP’s Mechanism Is Distinctive

Unlike conventional sleep compounds that act through a single sedative pathway, DSIP interacts with several overlapping neurochemical systems, making it a uniquely complex subject for research.

System Primary Research Effects
NMDA Receptors Modulates excitatory neurotransmission; may reduce excessive neural stimulation in cortical and hippocampal tissue
GABAergic Pathway Potentiates GABA-activated currents in hippocampal and cerebellar neurons, supporting inhibitory signaling
Opioid System Acts indirectly via stimulation of Met-enkephalin release; does not bind opioid receptors directly
HPA Axis Reduces basal corticotropin (ACTH) levels and attenuates stress-induced cortisol release
Pineal / Adrenergic Modulates α1-adrenergic receptor activity, influencing melatonin-related enzyme regulation

This multi-pathway profile is what distinguishes DSIP from simpler sleep peptides. Rather than forcing sedation through a single receptor, DSIP appears to modulate the neurochemical environment associated with slow-wave sleep initiation — while simultaneously influencing stress response systems that can interfere with sleep quality.

Research Applications

DSIP is used in studies examining:

  • Slow-wave (delta) sleep promotion and sleep architecture modulation
  • HPA axis regulation and stress-induced corticotropin dynamics
  • NMDA and GABA receptor interaction in cortical and hippocampal tissue models
  • Neuroprotection and mitochondrial function under oxidative stress conditions
  • Opioid and alcohol withdrawal symptom attenuation
  • Analgesic mechanisms and antinociceptive pathways
  • Motor recovery and stroke rehabilitation models
  • Circadian rhythm regulation and chronopharmacological programming

Specifications

Format Lyophilized powder
Purity ≥99%
Aliases Delta Sleep-Inducing Peptide, Deltaran
Available sizes 5mg
Storage 2–8°C unopened; stable 12+ months
Use Research purposes only — not for human use

Reconstitution

DSIP arrives as lyophilized powder and must be reconstituted with bacteriostatic water prior to use. Use the formula:

Total mg ÷ Volume added (mL) = Concentration (mg/mL)

Example: 5mg vial + 3mL BAC water = ~1.67mg/mL solution

Reconstituted peptide should be stored at 2–8°C, protected from light, and used within 28–30 days. Do not freeze reconstituted solution — freeze-thaw cycles degrade peptide structure and reduce potency.

Protocol Notes

DSIP operates at microgram-level doses, significantly lower than most research peptides. Administration is typically timed in the evening to align with natural sleep cycle initiation.

Typical research dosing framework:

  • Starting dose: 100–200mcg per administration
  • Escalation: Increase by 50–100mcg as needed based on observed response
  • Frequency: Once daily, typically 30–60 minutes before the intended sleep window
  • Study duration: 2–4 week cycles are common; longer protocols (8–12 weeks) have been used in stress and neuroendocrine research

Commonly observed effects in research models:

  • Sleep-related: Changes in slow-wave sleep duration and latency, REM suppression in some models — effects are variable across species and subject conditions
  • Neuroendocrine: Modulation of cortisol, ACTH, LH, and growth hormone-related secretion patterns
  • Other: Transient headache, nausea, or mild dizziness reported in human-subject studies at higher doses; injection site irritation; orthostatic blood pressure fluctuation at escalating doses

Research Stacks

DSIP is commonly paired in research settings with:

  • Selank — Selank influences GABAergic tone and enkephalin metabolism; it is studied alongside DSIP in sleep and stress-response models where overlapping effects on delta-wave induction and anxiolytic signaling are examined.
  • Semax — Semax upregulates BDNF and modulates HPA axis activity; researchers pair it with DSIP to assess how neurotrophin-promoting and sleep-regulatory peptides interact in neuroprotective and circadian research frameworks.
  • Epithalon — Epithalon influences pineal melatonin synthesis and telomerase activity; it is co-studied with DSIP in longevity and circadian biology models to examine convergent effects on sleep architecture and neuroendocrine aging markers.
